Evaluation of TNF α and TGF β polymorphism in Egyptian patients with Chronic HCV

Abstract


This study aimed to evaluate the association betweentumor necrosis factorα(TNFα) –G308A&Transforming growth factorβ(TGFβ) -C509T polymorphismsand different liver pathological changes in Egyptian patients infected with hepatitis C virus (HCV).This study included 122 subjects; 72 treatment-naïve chronic HCV patients with different fibrosis grades (F0-F4) and 50 healthy subjects. The TNFα −G308A& TGF –C509T regionswere amplified by PCR and the different genotypes were detected by restriction fragment length polymorphism (RFLP) analysis.
The TNFα& TGFβ proteins was detected by ELISA. The influence of different TNFα −G308A & TGF –C509T genotypes on TNFα & TGFβ expression and liver disease progression were statistically analyzed. The odds ratio (OR) and 95% confidence interval (CI) were calculated to assess the relative risk confidence.
The obtained results showed that TNF and TGF SNPs frequencies were significantly different between studied groups and associated with different levels of hepatic fibrosis from the obtained results data we conclude thatTNFα & TGFβ SNPs at nucleotide -308 & -509 respectively represents an important genetic marker that can be used for the prognosis of different liver pathological changes in HCV infected patients
