Abstract


This study was carried out in the laboratory of Plant Cellular and Molecular Genetics (PCMG), Agricultural Genetic Engineering Research Institute (AGERI), Agricultural Research Center (ARC), Ministry of Agriculture and Land Reclamation (MOALR), Giza, during the period from 1994 till 1998.
The aim • of the. present investigation was to study the initiation of plant tissue culture in lentil (Lens culinaris Medik.) cv. Precoz and Giza 370.
This work included two parts; the ftrst part deals with callus initiation and growth, while the second part deals with plant regeneration. The following factors were studied:
1. Cultivars.

2. Explant types
3. Growth regulators.
4. Media formulation

Part A. CaUus Initiation and Growth

1. Effect of 2,4-D concentrations, cultivars and explant typon callus induction and its characteristics
Studied the effect of three concentrations of2,4-D (5, 15 and
25 , cultivars and explant type on callus induction and its characteristics; i.e. callus inductionpercentage (CI%), callus fresh weight (CFW) after 15, 30 and 45 days from culturing (WO, WI and W2; respectively), callus growth value (CGV) between 15-30 and
30-40 days (A and B; respectively) and callus growth rate (CGR).

1.1.- Effect of2,4-D concentrations:
I. Increasing 2,4-D concentrations up to 25 J..M, callus induction percentage was decreased and this decreament was not differed significantly.
