A Study of the Effects of Clomiphene Citrateon the Histological Structure of the Fallopian Tube of Albino Rats: Possible Role of Isoflavone
Marwa Mohamed Safwat;
Abstract
Fallopian tube is a dynamic, steroid responsive tissue has fundamental roles in gamete transport, fertilization, and early embryo development. Clomiphene citrate (CC ) isnon steroidal synthetic hormone with estrogenic effect has been the first line therapy for induction of ovulation in women with anovulatory infertility.
The prolonged treatment of clomiphene citrate is associated with an increased frequency of tubal ectopic pregnancy in humans as,it causes destructive changes in the epithelium of the tube and block of oocyte.It was noticed that this damage was reversed with the administration of 17 beta-estradiol (E2).
Phytoestrogens are a group of non- steroidal compound of plant origin that present structural and functional similarities with17 β estradiol. Isoflavones, are the most widely known category. There are different mechanisms of action of isoflavones accepted, although they may be considered as selective modulators of estrogen receptor.
The goal of this work was todescribe the structural changes that occur in the oviduct after clomiphene citrate administration and the possible protective effect of isoflavone in restoring the integrity of the structure of the oviduct of prepubertal and adult albino rats when it was given with clomiphene citrate.
Forty-eight prepubertal and adult Female albino rats were chosen in the present study. The rats were divided into eight groups six rats for each group
Group A: twenty four prepubertal rats (2- 3 weeks) were further divided into four subgroups,
Group A1 (control group): the rats were left without any intervention. Group A2 The rats were treated orally by soy-derived isoflavone (iprivone 300 mg tablets dissolved in their drinking water) in a daily dose of 100mg/kg and examined after two weeks. Group A3 the rats received clomiphene citrate orally 10mg/kg/day for four days. GroupA4 the rats received clomiphene citrate (10mg/kg/day) and isoflavone (iprivone 100mg/kg/day) for four days concomitantly followd by isoflavone only till the end of two weeks.
