“The Effect of Platelet Rich Plasma as Preventive and Therapeutic Measures on Lipid Profile in Chronic Liver Diseased Rats”

Abstract
The main objective of this study was to elucidate the possible preventive and therapeutic role of Platelet Rich Plasma on lipid profile in Thioacetamide-induced chronic liver diseased rats.Sixty female albino rats divided into Six groups were allocated into the following groups:1) 4 weeks groups:I. control group: (n=10) received only normal saline/3 days and were studied by the end of the 4th week. II. Thioacetamide group (TAA group): (n=10) Rats of this group were subjected to intraperitoneal (i.p.) injection of TAA (200 mg/kg) every 3 days for 4 weeks for induction of liver cirrhosis.III. Thioacetamide - platelet rich plasma early treated group (TAA-PRP early group): (n=10)To clarify the possible protective effects of platelet rich plasma (PRP), rats of this group were given TAA (200 mg/kg) every 3 days followed by immediate administration of PRP and rats were studied by the end of the 4thweek.2)6 weeks groups: I. control group of rats: (n=8)received only normal saline/3 days and were studied by the end of the 6thweek.II.Thioacetamide group (TAA group): (n=11)Rats of this group were subjected to intraperitoneal injection of TAA (200mg/kg) every 3days for 4weeks and were studied by the end of 6th week.III. Thioacetamide - platelet rich plasma late treated group (TAA-PRP late group): (n=11)To clarify the possible therapeutic effects of platelet rich plasma (PRP), rats of this group were given TAA as previously described, administration of PRP was started after 2 weeks and for 4weeks and rats were studied by the end of the 6th week. The obtained results revealed that,a significant increase in serum levels of AST, ALT,MDA and liver MDA concentrations were observed in TAA injected rats. However, administration of PRP in TAA induced liver toxicity in rats exhibited a significant decreased in all mentioned parameters and attenuated the increased MDA level in liver tissues. Plasma lipid profile reduction were observed inTAA induced hepatic toxicity in rats when compared with control group. Meanwhile, PRP administrations resulted in significantincrease in all mentioned parameters. It could be concluded that, inhibitionof peroxidation, inflammation and oxidative stress and enhancedantioxidant status in rat liver tissues by PRP suggest the potential efficacy of PRPas an addition hepatoprotective and anti-inflammatory in treatment of chronic liver disease.
