THE EFFECT OF WHARTON’S JELLY MESENCHYMAL STEM CELL PRODUCTS AND L-AMINO ACID OXIDASE ON SQUAMOUS CELL CARCINOMA CELL LINE

Abstract


Head and neck squamous cell carcinoma (HNSCC) accounts for over 90% of all head and neck cancers. The umbilical cord is the cable linking the fetus to the placenta. It usually has two arteries and a vein, suspended in a hydrated extracellular matrix known as Wharton’s Jelly. Wharton’s jelly mesenchymal stem cells (WJMSCs) may be more useful for cancer therapy than other adult stem cells, since they are easily prepared in relatively large quantities from umbilical cord after delivery and pose no ethical issues. L-amino acid oxidases (LAAOs) are widely distributed in the venomous snake families Viperidae, Crotalidae and Elapidae. Several related proteins with LAAO activity have been described that exhibit antimicrobial, antineoplastic or apoptosis inducing activity. It is assumed that its general mechanism of toxicity is based on the generation of cytotoxic amounts of H2O2.
This study was carried out to determine the anticancer effects of WJMSCs conditioned medium (WJMSCs-CM), L-amino acid oxidase (LAAO) and the combination of WJMSCs-CM and LAAO on HEP-2 cell line after 24, 48 and 72 hours. Flowcytometry was used to detect the percentage of apoptosis and real time PCR was used to detect the expression of p53 and Bcl-2 genes.
The results of the study showed that LAAO alone had the greatest cytotoxic effect on HEP-2 cells followed by the combination between WJMSCs-CM and LAAO then WJMSCs-CM alone.
From the results of this study it could be concluded that the maximum effect of WJMSCs-CM and the combination on HEP-2 cells was noted after 48 hours but, the maximum effect of L-amino acid oxidase (LAAO) on HEP-2 cells was noted after 72 hours.
