Effect of Rofecoxib (A new selective COX II inhibitor) on plasma insulin level & pancreatic free hydroxyl radicals in diabetic rats.

Mohamad Mahmoud Abdel Rahim El Shaer;

Abstract

This work is a trial to postulate the pretreatment effects of a newly introduced selective COX-2 inhibitor namely
{Rofecoxib} versus a non-selective COX inhibitor namely

{Indomethacin} on plasma insulin levels and pancreatic free hydroxyl radicals as well as histopathological changes of islets beta cells in an experimental model of islet beta cell dysfunction in rats by streptozotocin.

Four groups of white male albino rats were used, each comprising twenty rats. 151 group served as control and the remaining three groups were treated with the test drugs and streptozotocin..

Experiments were done on streptozotocin treated rats to study the prophylactic effects of Rofecoxib versus Indomethacin on:

1). Plasma insulin levels both basal and half an hour after glucose load.
2). Histopathological changes of islet beta cells m different pancreatic sections.
3). MDA level in freshly prepared pancreatic homogenates
