Effect of Autoclave Sterilization on Cutting Efficiency of Two Different Single Rotary File Systems (An In Vitro Study)

Abstract


The aim of this study was to evaluate the effect of autoclave sterilization on One shape by comparing it with Twisted file regarding their cuttingefficiency which assessed by volumetric change in root canal as well as change in surface topography with sterilization cycles.
Volumetric change of root canal was assessed by comparing pre- and post- instrumentation measurements taken on CBCT images,while the change of surface topography was evaluated by scanning electron microscopy.
A total of sixty four human permanent mandibular molars were used in this study. Access cavities were done; and then special measures were taken to ensure standardization and reproducibility of CBCT images.
Samples were divided into two groups according to the file used in instrumentation; samples were prepared by One shape file (group ׀) and samples prepared by Twisted file (group ׀׀). Each group was further subdivided into four subgroups(subgroup A, subgroup B and subgroup C, subgroup D) according to the number of sterilization cycles which were subjected to. The teeth were fixed in position inside a block of acrylic resin during the whole study and then the teeth were imaged by CBCT before cleaning and shaping.
Cutting efficiency assessment using CBCT images analysis:
Change in canal volume was assessed by comparing pre- and post- CBCT images in the sagittal sections.Fusion of the pre-instrumentation and post-instrumentation records was achieved using special software that allows manual and then automatic registration of the primary and secondary images.Data were collected then statistical analysis was performed.2way anova was used to assess effect of file type and no of cycles over change in volume followed by tukey post hock test
Surface topography assessment using scanning electron microscopy:
Files were evaluated under SEM for defects in its new as well as used conditions to investigate effect of sterilization on the file surface topography.
Results:
Concerning cutting efficiency statistical analysis showed significant deferent increased in one shape file than twisted file, and with repeated sterilization a significant decrease in cutting efficiency regardless the type of file used.
Concerning surface topography scanning showed increase in surface roughness and deepening in irregularities.
Conclusion:
Under the conditions of the present study it can be concluded that:
1. Cutting efficiency of One shape file was higher than Twisted file.
2. Cutting efficiency of One shape file and Twisted file were affected by repeated sterilization cycles.
3. File surface topography was affected by repeated sterilization cycles.
