Molecular Genetics Studies of Some Developmental Abnormalities in Wild Mice

Abeer Fikry El-Sayed El-Nahas;

Abstract

The present work aims to throw light on the effect of the combination between one of Kit mutations (KitW} and other Kit alleles originated from wild mice as KitM allele from Japanese wild mice (MOG) or KitN allele from Mus m. musculus (NJL) on the fertility of these mice. This work based on the fact that, the combination between Kitw allele and Kits allele from Spanish wild mice (Mus spretus) lead to sterility of the males.

This study was carried out through: I. Congenic mice strain.
The mice were obtained and maintained as inbred strains at the

laboratory on Hokkaido University in Japan. The wild mice were used for construction of two-congenic mice strains using inbred laboratory mice (C57BL) as background.

However, this study was carried out by:

D. Simple sequence length polymorphism (SSLP):

For mapping of MOG and NJL Kit allele on chromosome 5 of C57BL. To detect which part of chromosomal region adjacent to Kit locus derived from MOG or NJL in the two-congenic strains.
m. Nucleotide sequence of Kit eDNA
III. a. From C57BL compared with that of Balb/C III. b. From MOG compared with that of C57BL.
