Abstract


Inflammatory bowel disease (IBD) is an idiopathic disease caused by a dysregulated immune response to host intestinal microflora. The two major types of IBD are ulcerative colitis (UC) which is limited to the colon and Crohn disease (CD) which can affect any segment of the gastrointestinal tract from the mouth to the anus.The clinical course is characterized by exacerbations and remissions and its main treatment depend on corticosteroids.The later have to bind to glucocorticoid receptors to exert their action (Da Silva et al., 2014 and Rowe et al., 2016).
Polymorphisms of Glucocorticoid Receptor (GR) gene lead to alternation in glucocorticoid sensitivity. Bcl1 is one of the most common gene polymorphisms under research in the past few years (Tadeusz et al., 2011 and Galecka E., 2013 and Prystupa et al., 2015).
The present study investigated the frequency of Bcl1 gene polymorphism in patients with IBD and evaluate its association with sensitivity to glucocorticoids therapy.
For this purpose, samples were collected from 50 patients diagnosed as IBD according to clinical, endoscopic and histopathological examinations (Group I) and 40 age and sex matched apparently healthy controls (Group II). Patients were subdivided according to the response to GC therapy into GCs responders and GCs non-responders.
All individuals in the study were subjected to full history including present history, family history and past history, general clinical examination, biopsy for site of the disease determination by sigmoidoscopy or full colonoscopy, general laboratory investigations including CBC, CRP and ESR and assay of Bcl1 gene polymorphism by PCR amplification and restriction analysis.
