Abstract
Attenuated RVF vaccme was prepared in BHK cells, then lyophilized. The original virus titre of reconstituted vaccine was I 08
• TCID50 I ml and after lyophilization, the titre of reconstituted vaccine was I 01 6 TCID50 I mi.
2. Evaluation of the lyophilized vaccine by testing its sterility, safety and potency. It was found that the vaccine was sterile, and safe when inoculated into hamster, sheep, cattle which remain alive for 2I days without any clinical symptom ofRVF disease, then the potency test was done by titration in BHK cell and titre was found to be log10 7.6
TCID5o I mi.
3. conduction of SNT on sera from vaccinated sheep after 28 day of vaccination. The neutralizing titre ranged between 40 and 80.
4. Physiological saline and distilled water were used as diluents for reconstitution of the lyophilized vaccine.
5. Various temperature degrees were selected for preserving either the
lyophilized or the reconstituted vaccine.
a. For the lyophilized vaccine, the selected temperature were 5'°C,
37°C, 25°C, +4°C, -20°C, -70°C), while for the reconstituted vaccine, the temperature of choice were (56°C, 37°C, 25°C, +4°C).
