Chemistry and Biology of Phenolics Isolated from Melaleuca leucadendra L. (Myrtacae)
A novel flavonol glucoside, Myricetin 3-O-β-4C1- galactopyranouronoide isolated from the aqueous ethanolic leaves extract of Melaleuca leucadendra besides, 15 known compounds were characterized as well. 11 of them were isolated for the first time from genus Melaleuca. The structures of all compounds were elucidated on the basis of spectroscopic data including 1D and 2D NMR and HRESIMS. On the other hand, the in vitro antioxidant activity using DPPH assay for the extract and new isolated compound 14 showed IC50 of 7.32±2.71 μg/ml and IC50 of 5.21± 0.87 μg/ml respectively. Also, using the oxygen radical absorbance capacity (ORAC) method the extract exhibited a free radical scavenger activity with an IC50 of 14.5 µg/ml. The lowest antioxidant capacity was found for fraction 1, with an IC50 value of 150.93 µg/ml. However, the results of all other fractions 2-9, and the tested isolate were in the range of 4-12 µg/ml the new natural product 14 with an IC50 value of 4.56 µg/ml. in addition, In vivo antioxidant activity using SOD and MPO assays showed good antioxidant activity of the crude extract by decreasing superoxide anion and hydrogen peroxide amount. Moreover, the cytotoxic activity of the extract on Huh-7, HCT-116 and MCF-7 cell lines using NRU assay showed a greater cytotoxic effect on HCT-116 cell line with IC50 83.08 ± 0.71.furthermore, evaluate the immunological activity of extract using phagocytic index method showed increasing in phagocytosis of bacteria by neutrophils accompanied with increasing of extract concentration.
