Detection and genetic characterization of Avian Influenza Virus in some Egyptian Governorates

Abstract


AI is a disease that causes a panic to the ranchers and people in contact with birds as it cause severe losses to investors in the field of poultry and death of human beings in contact with infected birds. Therefore this study were done on 167 samples from broilers, layers, breeders, ducks and pigeons in several governorates of Alexandria, Behera, Kafr El-Sheikh, Gharbia, Dakahlia, Menoufiya and Fayoum. These samples were used for the detection of avian influenza virus antigen in chickens in different governorates by immunochromatography, detection of avian influenza virus RNA in chickens by RT-PCR, studying sensitivity and specificity of immunochromatography in relation i:o RT-PCR for detection of avian influenza virus in chickens, sequencing and phylogenetic analysis of the local avian influenza virus. These results achieved the following:

The incidence of AI outbreaks diagnosed by RT-PCR in broiler, layer and breeder flocks were 59.6%, 45.5% and 50%, respectively.

AI virus was detected among broiler chickens by RT-PCR in Alexandria, Behera, Dakahlia and Gharbia governorates at a percentage of 53.3%, 73.4%, 50% and zero%, respectively in examined flocks. AI viral antigen was detected in broiler chickens by immunochromatography in Alexandria, Behera, Dakahlia and Gharbia governorates at a percentage of 40%, 20%, 25% and 100%, respectively in examined flocks.

The detection of AI virus in layers by RT-PCR indicated that incidence was 40% and 100% in Behera and Gharbia governorates, respectively in examined flocks. The virus could not be detected in examined layer in Alexandria governorate.

With regard to the results of detection of AI virus antigen by immunochromatography, the virus was detected in examined layers at a percentage of 50% and 100% in Behera and Gharbia governorate, respectively. On the other hand, the virus could not be detected among examined layer in Alexandria governorate.

AI virus was detected in broiler breeders by RT-PCR and immunochromatography in the examined samples in Behera and Fayoum and could not be detected in the examined samples in Alexandria and Gharbia governorates.
