Comparative Study of the Effect of Insulin and Green Tea on the Parotid Gland of Streptozotocin Induced Diabetic Rats (Histological and Electron Microscopic Study)

Abstract


The aim of this study was to compare between the effect of insulin and GT on the PG of STZ induced diabetic Albino rats histologically and ultrastructurally.
Forty male Albino rats (weighing between 200-250 gm each) were used in this study.
The animals were divided into the following groups:
1) Control group: (10 rats)
This group contained ten rats which received an equivalent volume of distilled water (solvent of GT) to that of green tea intragastrically for 17 days and a single dose of 1ml of citrate buffer (solvent of STZ) through the vein tail under anesthesia.
2)Experimental (Diabetic) group: (30 rats)
The rats have fasted for 14 h. and diabetes was induced by a single injection of 40mg/kg body weight STZ freshly dissolved in 1 ml Citrate buffer (0.01 M; pH 4.5) through the tail vein under ether anaesthesia. After the injection, the animals were given free access to water and food.
Blood samples were obtained via vein puncture of tail vein. A plasma glucose level greater than 300mg/dl confirmed the occurrence of diabetes that was determined 3 days after the drug injection using a glucometer .
Then the experimental group was divided into three subgroups:
a) Diabetic untreated subgroup:
It contained ten diabetic rats which were left untreated.
b) Insulin treated subgroup:
It contained ten rats which received a daily dose of insulin for two weeks (5 IU/kg body weight/day, Novo Industri.) after confirmation of diabetes subcutaneously. The rats received the last insulin dose 24 hours before being sacrificed.
c) Green tea treated subgroup:
It contained ten rats which received green tea daily in a dose of 1 ml/ 100 gm body weight for two weeks in the form of an aqueous extract at a concentration of 0.2 g/ml. This was obtained by soaking the green tea in boiling water and after 5 min the extract was filtered and the filtrate was fed intragastrically to the rats.

At the end of the experiment period, rats were sacrificed separately by overdose of anesthesia and their heads were immediately dissected to obtain the parotid salivary glands. The gland was excised and half of the specimens were prepared to be examined by light microscope and the other half was prepared and examined by transmission electron microscopic.
