In Vitro Detection of Anti-endothelial Cell Antibodies in Chronic Renal Failure Patients Before Transplantation

Rasha Mohy El-Din Badawi;

Abstract

Over the past decade, endothelial cells (I:Cs) have been
the subject of extensive investigation due to their multifaceted role in the development of the immune response

Transplantation of an organ results in din :ct exposure of endothelial cells to circulating blood, and hus a foreign environment in which various immune reactic ns are primary evoked in response to antigens expressed by end Jthelial cells.

Patients waiting for renal transplantation could develop HLA antibodies and also non-HLA antibodie: (AECA). The last group have antibodies directed against e 1dothelial cell antigens, and are negative for the classicc 1 cross match technique. However, they could develop vascula •rejection.

So, our goal was to provide estimate of• he presence of AECA in kidney transplant patients before t •ansplantation, since it is important to avoid the occurance ofk dney rejection pnor immunization and sensitization to antiger of endothelial cells.

Cross match tests were performed for a: l chronic renal failure patients. Sixty serum samples with 1egative cross match were collected from patients before tranplantation and stored at -20°C for the study.

ELISA plates were prepared for retrospe :::tive detection of antiendothelial cell antibodies.
