Abstract
s
results in a high titers of (109.16 , 108.8 and 108 5 EID50/ml ) with corresponding HA titer of (8log2) for the strain (A/Ch/PA/13609/93, A/Ty/MN/3689_1551/81 and A/Ty/CA/209092/02) in order.
Regarding the effect of the route of inoculation on virus titer and HA activity, results represented that the highest infectivity titer and HA activity were obtained after using the allantoic sac, the amniotic sac, CAM and finally the yolk sac routes
in order, however the allantoic sac route was found to be the best route for the

three used H5N2 AI virus to obtain high titers of 109•

and 10 • EID50 I ml
with corresponding HA activity of 8.5 log 2, 8 log 2 and 8 log 2 for the strains A/Ch/PA/13609/93, A/Ty/MN/3689_1551/81 and A/Ty/CA/209092/02 m descending order.
It was also noticed that the allantoic fluid followed by the chorioallantoic membrane, were the best sources of harvestation. The allantoic fluid recorded a

titer of 109•
, 10 •
and 10 • EID50/ml with the highest HA of 8 log2 for mentioned
viruses respectively. And the recorded titers obtained from the chorioallantoic membrane by three successive freezing and thawing were 10 8,108 3 and 1 08.4EID
50 /ml. for the mentioned used viruses respectively, however the corresponding HA activity by the chorioallantoic membrane for the first two viruses was 8 log2 where as it was 5 log2 for the 3rd strain A/TY/CA209092/02.
Concerning the determination of the optimal time for harvesting the virus yield, result clarified that a high infectivity titers were obtained starting at 60 hrs and ended 72 hrs, the recorded highest titers for strain A/Ch/PA/13609/93 were
