HE4 Expression in the Ovarian Malignancy and its Relation to the Risk of Malignancy Index (RMI)

Abstract


This study was done at Medical Biochemistry Department, Ain Shams University Faculty of Medicine during the period from 2013 – 2014 and included 50 female patients with ovarian tumors who were admitted to AinShams University Maternity Hospital, gynecologic outpatient clinic.
The aim of this study was to ascertain HE4 (Human Epididymis Protein 4) clinical utility, to do a comprehensive assessment of HE4 protein expression in benign and malignant ovarian tissues by real time PCR and its relation to RMI (Risk of Malignancy Index) that may have predictive and therapeutic impact on these patient and to correlate HE4 expression with RMI to assess the diagnostic significance of this marker in malignant ovarian masses.
The studied individuals were classified into two main groups:
 Group A: Patients with malignant ovarian lesions. (n= 25).
 Group B: Patients with benign ovarian lesions as a control group (n=25).
All patients in our study were subjected to complete detailed history taking, general and local examination, routine laboratory investigations, and radio diagnostic investigations as pelvic US. Also CA125 levels were measuredand RMI were calculated.The ovarian masses or cysts tissues were further investigated byhistopathological evaluation after surgical removal, RNA extractionof all samples was done forrelative quantitative real time PCRof HE4 and β actin as a house keeping gene for both tissue samples. Then the results were calculated and statistically analyzed by the SPSS software.
