Role of Caloric Restriction Against Oxidative and Perfusion Challenges to• Isolated Hearts
Amira Metwally Abdel Rhman;
Abstract
This study was performed to clarify the cardioprotective action of food restriction against the myocardial injury and dysfucntion induced by oxidative stress and deperfusion-reperfusion of the isolated heart. The food-restricted regimen was achieved by an alternate day feeding method.
Food restricted rats were supplied by the same daily amount and composition of diet offered to their controls but in an alternate way, one day of feeding followed by one day of fasting and so on. The hearts isolated from food restricted rats and their matching controls were divided into three groups. Group I: deperfusion - reperfusion model: the isolated hearts were subjected to global ischemia for 30 min followed by a similar period of reperfusion. Group II: the hearts were perfused :with K-H bicarb. buffer solution at first for 15 min then a solution of H202 as a free radical
generating agent is used in a dose of 600to perfuse
hearts for 30 minutes. Group III: the isolated hearts were perfused at first by K.H bicarbonate buffer solution then perfused with H202 (in a dose of600
for 5 min then the hearts were subjected to global
ischemia for 30 min followed by a similar period of .. reperfusion.
