Abstract


Anti-K is the most common immune red cell antibody outside the ABO and Rh systems. Kell antibodies should be considered of clinical significant, causing severe HDN and HTRs. We described, RFLP-PCR assay using restriction enzyme Bsm I that recognizes the Kl nucleotid substitution. From the collected samples, 50 samples are chosen for RFLP-PCR genotyping. In all cases the results of analysis of Kell genotypes from P.B, C.B, A.F DNA samples agreed with the red blood cell Kell phenotypes. We concluded that, the application of the PCR for detennination of red cell blood genotype is a highly sensitive technique, less hazardous and reliable. We recommend; routine identification of women's Kell status the same as with Rh-typing using serological method
& determination of the fetal Kl and K2 genotype provides accurate and
timely information that can aid the prenatal care of women sensitized to these Kell antigens, and enable all pregnancies with Kell negative fetuses to be excluded from further follow up and further invasive assessments.
