Abstract

The present study has been carried out in Ain Shams Center for Genetic Engineering and Biotechnology Branch of Faculty of Agriculture(ACGEBFAC). Genetic characterization by biochemical and molecular techniques were carried out on the collected blood samples from three native Egyptian breeds: The two lines of Fayoumi breed ; GG line and PP line, Dandarawi breed and Doki-4 strain. The native protein gel electrophoresis was used to identify the biochemical genetic fingerprinting of each breed and produced 24-28 bands on the electrophoregram. Studying the follows : • Phenotypes (banding pattern) of the nine plasma protein loci. •The homogeneity of each breed. • The allelic frequencies along with detecting the biochemical genetic markers to differentiate
- within and between those breeds . Meanwhile, the molecular fingerprints
ii
has been identified by RAPD analysis using 10 primers (A16, A17, Al8, Bo8, B9, B16, CS, ClS, ubc30 and ubc75 ). That were successful in amplification of DNA and produced specific bands. These bands could be used as markers to characterize such breeds. The dendrogram was constructed to find the relationships between the breeds based on both biochemical and molecular analysis.
Key words: Molecular fingerprinting, Biochemical fingerprinting, Genetic markers, RAPD analysis, Allele frequencies, Dendrogram,
electrophoregram.
