Taxonomic and genetic studies of plants in Petrified Forest area of Egypt for documentation

Abstract


All plants in Petrified Forest protected Area of Egypt were collected during three seasons in 2014, 2015 and 2016.
The plants were collected in vegetative stages and flowering stages, to studied morphology, anatomically and genetically by plant DNA barcode. Macromorphological studies
by using different taxonomical keys. The identification was confirmed and authenticated in a herbarium at Faculty of Science Ain Shamus University
In the present study, 26 taxa belong to 14 families of dicotyledonous plants and 3 taxa belong to two families of monocotyledonous plants were collected from petrified forest protected area of Egypt during three flowering seasons 2014, 2015 and 2016. Morphologically, the results obtained show 20 taxa are herb, 5 taxa are sub shrub, 2 taxa shrub and two taxa (zilla spinose subsp spinose and Lycium shawii) spiny shrub.
Stem range from 24 Taxa are erect, two taxa (Erodium ciconium, Mesembryanthemum nodiflorum) are procumbent, one taxa (Pancratium sickenbergeri) bulb and one taxa (Phragmites austoralis) rhizome. leaf Type was 19 Taxa are simple leaf, 8 taxa lobed and two taxa (Trigonella stellate, Zygophyllum coccenium) are compound.

In this study, we tested the efficiency of three DNA barcoding loci located in chloroplast rbcL, matK & trnh-psbA and one loci located in nucleus ITS2 in identifying the plant species in the Petrified Forest Protected Area.
The success rate of PCR amplification was variable for all barcoding loci. The result indicates that using matK have very low PCR amplification success, only four plants from tested species (14 %).
In our study, the best PCR success rate was in ITS2 locus with 79 % of samples and high quality of sequencing success and variable enough to discriminate between species at species level so ITS2 is an efficient barcoding locus for identifying plant species and recently suggested by CBOL 2009 as an additional locus.
