The prophylactic action of vitamin B12 (HYDROXYCOBALAMIN) on the ultrastructure of the heptoytes of albino mice injected with the NSAID Diclofenac sodium Rheumarene .

Abstract

The NSAIDs are useful agents for treatment of rheumatic diseases and other inflammatory disorders .Nontheless ,in spite of the beneficial role of these drugs , they are incriminated of in producing certain pathological side effects on orther body organs .In this concern,few reports referred to the prophylactic effects of vitamin B12 or hydroxycobalamin against the dagmaging effects of these drugs or any other chemical agents , besides its known use for growth , cell reproduction , hematopoiesis , nucleoprotein and myelin synthesis .Thus the present investigation aimed to examine the prophylactic action of inttramuscular injection of vitamin B12 on the ultrastructure of the hepatocytes of albino mice treated with the NSAID DICLOFENAC sodium (Rheumarene).The administration of therapeutic dose of vitamin B12 induced mild changes in the ultrastructure of the hepatocytes including degeneration of the mitochondria , proliferation of cisternae of RER and marked aggregation of lipid droplets and glycogen besides congestion of the hepatic sinusoids .Injection with the NSAID Rheumarene caused severe mitochondrial devestations with obvious loss of their internal ridges or cristae and matrices .fragmentation and degranulation of RER as well dilated sinusoids and activated Kupffer cells .nonethless, pretreatment with the therapeutic dose of vitamin B12 for 10 days induced prophylactic activity against the parhogenic impacts of the NSAID rheumarene in the hepatocytes which appeared more or less normal except for very minor abnormalities of the mitochondria ,RER OF the hepatocytes and the hepatic sinusoids .
