STUDY ON GROWTH CURVE OF BOVINE CORONAVIRUS (BCV) ON MDBK CEll CULTURE.

SAMIA SAID ABD EL-NABI;

Abstract

Neonatal calf diarrhoea affecting calves during the first few weeks after birth causing major economic losses directly through mortalities and indirectly from poor growth after clinical disease. Studies throughout the world have establishedthe important role of BCV, BRC as well as k99 producing enterotoxigenic E. coli in the disease complex.

Under optimum condition of temperature, pH and trypsin concentration as well as absence of serum in maintenance medium Coronavirus growth characteristics were evaluated.

The study was planned to follow the growth curve of Bovine Coronavirus and its characteristics as a step on the way of production of vaccine also to assess the following :

• Kinetic growth characteristic of bovine Coronavirus in suitable virus host system normally MDBK cells.
• Sequential follow up of bovine Coronavirus protein biosynthesis and its antigenic property.
The results of this study revealed the following :

• The virus was propagated on MDBK cells 3 times till clear CPE for

Coronavirus was obtained.

• The CPE was started to appear after 24 hours post inoculation and reached maximum level at 72 hours after that the sheet showed cavitation at 96 hours post inoculation and complete destruction of
cell sheet at 120 hours P.I.
