P153 AND Kl-67 EXPRESSION IN SOME PROLIFERATIVE BENIGN AND MALIGNANT SKIN LESIONS
ELHAM ABDEL-AZIZ SARHAN;
Abstract
P53 is a tumour suppressor gene which regulates the cell growth. It
1s located on chromosome 17 P and encodes a 53 Kd phosphoprotein. This PS3 protein exerts important biological activities in various normal
and pathological human tissues.
..,
'
P53 in its natural form or "wild type" can bind to DNA and prevent cells from entering into the S-phase ofthe cell cycle. This wild type P53 acts as a potent suppressor of uncontrolled cell division.
P53 is present in the nucleus of' all mammalian cells, usually at a concentration below the detection level for immunohistochemical methods, mainly due to its short half-life. In contrast mutation of P53 gene produces a P53 protein with a prolonged half-life that may reach a concentration in the cells that allows immunohistochemichal detection. Mutant P53 gene has the ability to transform cells and lead to tumorigenesi's.
P53 IS frequently mutated in human cancers especialy non melanoma skin cancers. But there is marked controversy about its role in benign hyperproliferative skin disorders.
• Ki-67 which is a nuclear antigen 1s a reliable biomarker for the detection ofproliferating cells in tissue samples.
The aim of this study was to identify the expression ofP53 protein and Ki--67 in psoriatic skin as well as in BCC and SCC. Also to detect DNA ploidy of cells in these diseases.
