Molecular and physiological studies on transformation of Vicia faba L. to enhance tolerance to salt stress
Hanan Samir Salah El- Din Ali;
Abstract
Arabidopsis thaliana AtNHX1 was previously identified as an important salt tolerance gene that transports Na+ into vacuoles. In the present study, AtNHX1 was transferred into the cells of the shoot apices of three Egyptian faba bean (Vicia faba L.) cultivars (Egypt 1, Giza 40 and Giza 843) via Agrobacterium-mediated transformation. Transgenic V. faba plantlets containing AtNHX1 and the selective marker encoding bialophos resistance (bar) were confirmed using (PCR). The expression of AtNHX1 in transgenic plants was confirmed by real-time PCR and western blot which indicated that the gene was highly expressed in transgenic plants.
