Assessment of possible role of berberine against-induced colon carcinoma in experimental rats

Abstract


The present work aimed to study the possible protective effect ofberberine against 1.2–dimethylhydrazine induced colon carcinogenesis in experimental animal.To evaluate the possible role of berberine in vivo, forty male Swiss albino rats randomly divided into four equal groups, ten animals each, as follows:
Group I: Rats kept as controls received daily 1 ml of physiological saline by gastric gavage.
Group II: Rats received daily 75 mg BRR/Kg b.w.(orally) by gastric gavage twice a week for 8 consecutive weeks.
Group III: Rats were injected with 20 mg DMH/kg b.w.subcutaneous once a week for 8 consecutive weeks.
Group IV: Rats received 75 mg BRR/Kg b.w. by gastric gavage (as a preventive treatment) for 15 days before DMH intoxication and concurrently with DMH over 8 weeks.At the end of experiment, Rats were sacrificed, and tumor tissues were dissected.
The result revealed that injection of DMH decreased the antioxidants levels (GSH and SOD) and increased inflammatory markers (MPO, MAPK and COX-2).More over it down regulated apoptotic markers (Caspase-3 and P53) expression that confirmed bycolon cell proliferation.The prophylactic effect of BRR was noticed as its administration increased antioxidants status and apoptotic markers expression that associated with inflammatory markers down-regulation with absence of proliferated colon cells.Also histological findings revealed that control group showed normal
