Trials For Freezing Of Buffalo Semen For Maximal Viability And Acrosomal Integrity
Amal Hassan Ali;
Abstract
The present study aimed at optimizing the equilibration time , the cooling regime and the adaptation of buffalo spermatozoa to 5C during
•. • processing of buffalo semen.
1-Glycerolization at 5°C was performed to clarify the effect of different cooling regimes. 1, 1 Y:z, 2hr), adaptation times. 1, and 1 Y:z hr), and equilibration periods (0, 1, 2, and 4 hr) on semen characters. On testing the effect of different cooling regimes adaptation time hr) and equilibration period (2 hr) were fixed, while n testing the effect of different adaptation times the cooling timehr) and equilibration period were fixed and on testing the effect of different equilibration periods the cooling timehr) and adaptation timehr) were fixed.
Eight adult 6-8 years old buffalo bulls were used in the current
study. 3 semen diluents (Milk, Tris, and sodium citrate) were used in the
• experiment. Pooled semen sample was divided into 3 portions which
diluted partially with the non glycerolized part of the used diluents. The
. diluted semen samples were gradually cooled and adapted at 5°C at which the second glycerolized portion of the diluents were added and
. kept for equilibration time, and after that the equilibrated semen samples
were exposed to the classical freezing process and kept in deep liquid nitrogen tank for at least 24 hrs. The individual motility, and acrosomal integrity were tested in each.' step of processing , and the viability index was calculated after thawing..
