EFFECT OF VITAMIN E 11AN ANTIOXIDANT" AND DEFEROXAMINE 11AN IRON CHELATOR" ON GENTAMICIN-INDUCED NEPHROTOXICITY IN ALBINO RATS

Abstract

Aminoglycosides, primarily gentamicin, are the most commonly used antibiotics worldwide despite their toxicity to the kidney and the inner ear. A preventive therapy against these side effects should combine safety and efficacy with low cost.

The approach was based on the novel observations that aminoglycoside antibiotic form a complex with iron and are capable of catalyzing the formation of free radicals in vitro and in intact cells. This mechanism may also be the basis of free radical generation by aminoglycosides observed in isolated mitochondria.

The aim of the present study was to evaluate the effect of vitamin E (alpha tocopherol) as an antioxidant and deferoxamine as an iron chelator has antioxidant property on nephrotoxicity induced by gentamicin. This study tried to find a design of protective therapy for gentamicin nephrotoxicity in rats, which are the suitable models of experimental animals for studying of gentamicin nephrotoxicity in humans.
