EFFECT OF SOME VASOACTIVE AGENTS ON VASCULAR REACTIVITY OF ISOLATED PORTO-MESENTERIC VEIN IN LIVER CIRRHOTIC RATS

Abstract

The effect of propranolol or somatostatin analogue (octreotide), in addition the possible role of prostaglandins •and serotonin on blood flow velocity and vascular reactivity of portal vein of liver cirrhotic rats were investigated. Sixty . eight adult albino rats of both sexes of local strains were included. Rats were divided into two main groups after a period of acclimatization. Group I (n=10) received nonnal saline and served as control non-cirrhotic group. Group II (n=;:58) in which liver cirrhosis was experimentally indticed using

CC14 (in a. dose of lml/kg by orogastric tube twice/week for ten weeks), after hepatic microsomal enzymes sensitization period of two weeks using phenobarbitone (350 mg/L. in their drinking water daily). . ' . . . Eight rats of the CC14 treated group were.chosenrandomly and their liver were examined histopathlogically to confirm liver cirrhosis. After induction ofliver cirrhosis, cirrhotic rats (50) were subdevided

into five subequal grmtps. Group llA injected intramuscularly with lml . . . distilled water/day for two successive weeks and served as cirrhotic non treated rats. Group. liB . rats were injected intramuscularly. daily for two successive• weeks with propranolol in a dose of (0.33 mg/kg). Group IIC • rats were injected subcutaneously daily for two successive weeks with octreotide in •a dose • of (100 • Lg/kg). Group IID rats• were injected with aspirin intramuscularly daily for two successive weeks in a dose of (1 00 ing/kg). Group liE ratswere injected with ritanserin intramuscularly daily for two successive weeks in a dose of (0.63 mg/kg).
