FURTHER STUDIES ON PIGEON PARAMYXOVIRUS VACCINE

Abstract

This study was done on the Preparation of different types of inactivated PPMV -1 vaccine using a varitey of inacti vators and adjuvants. At the same time a trials for atttinuation of the local Egyptian PPMV-1 isolate was done.

The preparecd types of vaccines were subjected to vaccine evaluation protocal including sterility, safety and potency as well as serological tests. The attinuated local isolate was subjected to evaluation by potency tests, serological test and histopathological investigation.

1- Passages of Master seed Virus :

Results show that the infectivity titre as well as the HA titre in creased by passaging the viruse in SPF eggs till fixed and reached the highest

titre by the 3rd passage and it was noticed that the highest virus titre obtained when the inoculum dilution was I 06 EID50, the most preferable day for harvestation was the 4th day and harvested CAF containe maximum virus titre than the embryo tissues.

2- Propagation of PPMV-1 in cell culture :

Studing the growth Curve of PPMV-1 on VERO and CER eel culture showed that the initial production of virus progeny occured 8 hours and 6 hours post infection for VERO and CER eels respectively. Passaging of the virus on CER, VERO and CEF cell culture showed noticible in crease in virus titre.
