Study On Trial For Preparation Of Fowl Pox Vaccine Locally On Embryonated Specific Pathogen Free Eggs

NAMAA ABD EL-AZIZ ABD ELHAFIZ MOHAMED;

Abstract

This study was planned to propagate the fowl pox virus (WPI)
I vaccinal strain in SPF embryonated chicken eggs and primary embryo fibroblast cell culture (CEF) for obtaining an efficient local vaccine.
I

I Pathogenicity assay of the original egg adapted FPV strain (WPI) in• ECE revealed titres of IOis EID50/ml and I 06•2 TCID50/ml in ECE and CEF
I cell culture, respectively.

I Growth rate of the WP I strain was studied, titration of inoculated I CEF cell culture and embryonated egg was carried out to investigate the best time for virus harvestation. Our results revealed that maximum titre of
I inoculated culture was around 120th hours post inoculation and on the 4th
I day after ECE inoculation.

I Six batches was successively prepared from. each vaccine type.
Sterility tests was carried out to evaluate the two prepared vaccines. After examination for any contaminants, the vaccines lyophilized by addition of
I sucrose-lactalbumin stabilizer, then repeated testing of sterility.
11 Titration was carried out, there was a gradual adaptation of the virus
in ECE cell culture accompanied by an increase in the virus titre. The highest titre batches (107 6 EID50 I ml for 5th batch of egg propagated
