The effect of Epitranscriptomic writer (METTL3) silencing on Hepatocellular carcinoma cell line

Afaf Rabea El sherbeni;

Abstract

Hepatocellular carcinoma (HCC) represents the sixth and fourth most prevalent cancer in worldwide and Egypt. Although the causal risk factors of HCC are well known; the fundamental molecular mechanisms are still principally indescribable. In liver carcinogenesis, epigenetic deregulations play a critical role. Reversible chemical modifications of mRNA have recently been recognized as essential regulatory mechanisms for regulation of gene expression, like DNA and core histone proteins.
METTL3 is an RNA methyltransferase responsible for N6-methyladenosine (m6A) modification of mRNA. METTL3 facilitates the translation of a diversity of oncogenes mRNAs implicated in the human carcinogenic process. On the other hand, METTL3 can epigenetically silence tumor suppressive genes via certain m6A readers - mediated mRNAs decay.
