Comparative pathological study on the anti-inflammatory effect of prednisolone and myrrh extract in a model of autoimmune disease using silica

Abstract


This study aimed to investigate the role of silica in the induction of autoimmune diseases in rats and developing an autoimmune disease model as well as investigate the therapeutic effect of myrrh in this autoimmune model compared to conventional treatment; prednisolone.
The autoimmune disease model was induced in adult female albino rats by different doses and routes of administration of crystalline sodium silicate suspension (5mg subcutaneously and orally and 7mg orally) once weekly for 20 weeks, then the rats were treated either with 500 mg/kg myrrh extract or 10 mg/kg prednisolone or with both orally for 6 weeks daily. Samples were collected at 20, 23, and 26 weeks of the experiment. All rats were euthanized at the end of the experiment by cervical dislocation.
To follow up on the development of the autoimmune disease, detection of anti-nuclear antibodies (ANA), gelatinases and collagenases (MMP-2, MMP-9 & MMP13), AST, ALT, BUN, and creatinine in the serum of euthanized rats was performed. Furthermore, oxidative stress biomarkers activity, TNF-α, and Bcl-2 gene expression were measured in the kidney, liver, and brain tissues. Histopathological examination of lung, kidney, liver, brain, and heart tissue was carried out. To investigate the anti-inflammatory effect of treatment agents in this study, histopathological examination of kidney and liver tissue and immunohistochemistry of TNF-α in the renal tissue was carried out.
Regarding Autoantibodies analysis result, all silica treated groups showed a significant increase in ANA at the 20th week with the highest increase in the group treated subcutaneously with crystalline silica.
The activity of gelatinases (MMP-2, MMP-9) and collagenases (MMP-13) increased in all silica exposed groups with a significant increase in the group treated subcutaneously with 5mg crystalline silica and group treated orally with 7mg crystalline silica in comparison to the control negative group.
