Potential impact of manganese nanoparticles in the management of hepatocellular carcinoma induced in rats

Abstract


Guided treatments with nanoparticles are a new approach in cancer therapy. This study aimed to evaluate the antitumor activity of manganese nanoparticles in vitro and in vivo in the management of hepatocellular carcinoma induced in rats. Manganese nanoparticles were biologically synthesized using Lactobacillus helveticus cells. MnNPs were characterized by means of transmission electron microscopy TEM and Dynamic light scattering DLS revealed that, MnNPs nanoparticles are of size range between 32.67-190.1 nm, Fourier transforms infrared spectroscopy ( FTIR ) that showed shifting in wave number of functional groups due to their involvement in MnNPs capping and synthesis. Ultraviolet-visible absorption MnNPs spectrum absorbance was analyzed by ultraviolet-visible (UV-VIS) spectrophotometer produced the absorbance spectra were scanned at 265nm. In vitro study of Manganese nanoparticles on HepG2 revealed IC50 of 41.5mM. The measurements were used to evaluate the efficiency of manganese nanoparticles, such as superoxide dismutase activity SOD and malondialdehyde level MDA , liver and kidney function tests, in addition to the evaluation of serum alpha-fetoprotein, caspase-3 and Heat shock protein -70 (HSP-70) in liver tissue. Total RNA extraction from liver tissue samples was also done for matrix metallopeptidase-9. Histopathological examination was also performed for liver tissue.
Results showed that manganese nanoparticles are effective in treatment of hepatocellular carcinoma induced by diethylnitrosamine in rats as it ameliorated the investigated parameters toward normal control animals. These findings were well appreciated with histopathological studies of diethylnitrosamine group treated with manganese nanoparticles, suggesting that manganese nanoparticles can serve as a
