Abstract
VI-Summary
Gout is a disease affecting human characterized by abnormal high level of serum uric acid lead to the formation and deposition of urate crystals in the joints and other tissues causing painful inflammation.
The present study was conducted to investigate the effect of natural safe source plants on the hyperuricemia, on the gene expression of xanthine oxidase enzyme and xanthine oxidase enzyme activity in rats. For realizing this purpose, 50 male albino rats were categorized into 5 equal groups as follows:
(GI) Control group (-ve control): non hyperuricemic normal rats.
(GII) Hyperuricemic group (+ve control): injected with potassium oxonate sol. i.p (250 mg/kg/ b wt.) once daily for consecutive 4 weeks.
(GIII) Febuxostat Hyperuricemic group: which injected with potassium oxonate sol. ip (250 mg/kg/ b wt.) then after 1 hour injected with Febuxostat sol. i.p (35 mg/kg/ b wt.) once daily for consecutive 4 weeks.
(GIV) Gingko biloba hyperuricemic group: which injected with potassium Oxonate sol. i.p (250 mg/kg/ b wt.) then after 1 hour injected gingko biloba extract. ip (80 mg/kg/ b wt.) once daily for consecutive 4 weeks.
(GV) Onion juice hyperuricemic group: which injected with potassium oxonate sol. i.p (250 mg/kg/ b wt.) then after 1 hour orally given onion juice (50 %) by gastric gavage once daily for consecutive 4 weeks.
At the end of experiment after 4 weeks, blood samples were collected from each rat in different group .The collected samples were centrifuged at 3000 rpm for 10 minutes to separate serum, the serum was employed for determination of xanthine oxidase, ALT, AST, ALP activities and uric acid, urea, creatinine and total protein concentrations.
After slaughtering of rats’ liver and kidney specimens were obtained and divided into:
I- Part one for estimation of MDA, GSH, uric acid contents and SOD, GSH-Px activities .
II- Part two was kept in 10% neutral buffered formalin for histopathological examination.
III- Part three (liver only) for studying xanthine oxidase gene expression.
