“Comparative study between the effect of Mesenchymal Stem Cells and/or Platelet-rich Plasma on Bone parameters in Ovariectomized mature Rats”

Arwa Abd El Moniem Hassan Ali;

Abstract

he present study was planned to elucidate the role of bone marrow derived mesenchymal MSCs (BM-MSCs) or platelet-rich plasma (PRP) and the combination of both in improving bone formation and slowing down bone loss and also to highlight the effect on bone markers in ovariectomized adult rats with osteoporosis.
The study was performed on 78 adult female rats, which were allocated into 5 groups:
Group I: Sham-operated control group (Sham) (n=16)
Rats were exposed to all surgical procedures without removal of the ovaries, and then after 4 weeks injected with 0.5 mL phosphate buffered saline (PBS) once i.v.

Group II: Ovariectomized (OVX) untreated group (n=15)
Rats were ovariectomized and then after 4 weeks injected with 0.5 ml PBS, once i.v.
Group III: Ovariectomized-MSCs (OVX-MSCs) treated group (n=14)
Rats were ovariectomized and, after 4 weeks, injected with rat bone marrow mesenchymal stem cells (BM-MSCs, 1×106) suspended in 0.5 mL PBS once i.v. (rat tail vein), using a modified protocol of Sui et al. (2016), and then taken for analysis 4 weeks later.
