Production and evaluation of antitumor efficiency of L-glutaminase enzyme from Egyptian fungal strains

Abstract


Nine fungal strains were screened by using a modified Czapek Dox's medium and PDA. The results indicated that Aspergillus fumigatus MT071344 was the most active fungal strain has the ability to produce L-glutaminase with activity 82 U/ml.
Ten natural wastes rich in glutamine such as rice straw, wheat straw, sugar cane bagasse, vermiculite, rice bran, wheat bran, soy bean, crushed corn, peanut peel and wheat grain were screened to choose the most potent one for optimum production of L-glutaminase enzyme. Soybean was found to be the best one for enzyme production with activity 72.5 U/ml.
A series of optimization processes was performed using DOE tools to detect the most potential conditions for the enzyme activity and production. The CCD and OFAT designs were used for screening important factors to improve and increase the production of L- glutaminase under SSF
