Development of Lateral flow immunochromatographic kits for rapid detection of Pasteurella multocida and Mannheimia hemolytica from cattle suffering respiratory infections
Mai Anwar Abd El-Samea;
Abstract
This study was undertaken to estimate the most antigenic part of P. multocida and M.hemolytica that can enhance immne response and result in production of specific antibody Also it was planned to develop and determine the diagnostic efficacy of P.multoida and M.hemolytica -specific lateral flow immunochromatographic kits for rapid, simple and onsite detection of P.multocida and M.hemolytica infection in clinical specimens for rapid detection of causative agents of BRDC. In the first part of the study standard strain of P. multocida and M.hemolytica taken from microbiology department faculty of VET medicine
These strain were examined bacteriologically for identification of P.multocida and M.hemolytica . The bacteriological examination indicated that strain is field strain of P.multocida and M.hemolytica. In the second part of the study is preparation of specific antigen of P.multocida and M.hemolytica (OMPs), immunization of rabbit with these antigen and also evaluation of immune response against this antigen and evaluation of antibody titer in the third part, a P.multocida and M.hemolytica -specific LFI kits was developed and its sensitivity, specificity and accuracy in detection of P.multocida, as compared to the standard bacteriological examination, reached to 88.2%, 92.4% and 91%, respectively and M.hemolytica reached to. 80.6%, 94.2% and 90%
