Abstract
yocardial infarction is considered to be one of the main causes of death from cardiovascular disease. The possible role of stem cells in the regeneration of ISO induced myocardial infarction was investigated. Telocytes are newly discovered cells, whose functions are not yet known. They had been suggested to have tandem with stem cells.
Eighty male Wistar albino rats were used in this experiment. Twenty young rats were used as a source of BM-MSCs. Sixty adult rats (180- 200gm) divided equally into three main groups:
Group I (Control group): rats were injected with 0.5 ml saline subcutaneously for 2 consecutive days and were equally divided into 4 subgroups (IA, IB, IC and ID). The rats were sacrificed after 3 days, 1 week, 2 weeks & 4 weeks, respectively.
Group II (MI group): rats served as a myocardial infarction model. The rats received isoprenaline hydrochloride (180mg/kg/day) for 2 consecutive days. The rats were subdivided into four equal subgroups (IIA, IIB, IIC and IID). The rats were sacrificed on the 3rd day, 1 week, 2 weeks and 4 weeks after induction of MI, respectively.
Group III (BM-MSC group): rats received ISO hydrochloride as in group II. Then, the rats were injected with BM-MSCs in the tail vein, 24 hours after the second dose of isoprenaline hydrochloride. The rats were subdivided into four subgroups (IIIA, IIIB, IIIC and IIID); 5 rats each. The rats were sacrificed on the 3rd day, 1 week, 2 weeks and 4 weeks after induction of MI, respectively.
In the current study, stem cells were isolated from the BM and then cultured for nine days. Examination of the culture showed that the BM-MSCs were mostly spindle -shaped, star shaped or fibroblast like appearance. By Giemsa stain, the adherent cells revealed bluish granular cytoplasm and multiple interdigitating processes.
Bone marrow MSCs were characterized by a positive immunostaining reaction for CD44 and CD105 and a negative reaction for CD34 and CD 19 by using flow cytometry.
Blood samples were taken on the third day of the experiment to measure the levels of troponin I and CKMB markers to confirm MI. Specimens from the left ventricles of the hearts of all groups were processed for light and electron microscopic examination.
Sections were stained by H&E, toluidine blue and Masson’s Trichrome stains. Immunohistochemical staining for anti CD 117, CD 34, CD 44, and anti VEGF antibodies was also performed.
