GENETICAL STUDIES ON SALT TOLERANCE, OF WHEAT ( T. aestivum L.) USING EMBRYO CULTURE TECHNIQUE

Ehab Mohamed Rabie Metwali;

Abstract

This investigation was carried out in the tissue culture laboratory and green house of the experimental form of the Faculty of Agriculture Suez Canal University, Ismailia, Egypt, during 1995 to 1998 years. The aim of the present study was detect the mechanisms of embryo culture initiation, regeneration and maintenance of different parental wheat genotypes and their hybrids under normal and sea water saline. Also, the nature of gene action and the genetic behaviour of callus growth and chemical component traits under laboratory conditions and yield components under green house conditions were performed through half diallel cross analysisincluding five parental genotypes under both saline and non-saline treatments. Also, screening both peroxidase and esterase isozyme polymorphism and extracted from call.us after sea salt treatments. Zymograms of both isozyme systems were obtained using discontinous - disscocicating PAGE technique.
