EVALUATION OF NANO-PARTICLES TRANSFER OF BIO-CONTROL RELATED GENE(S) VIAGENE EXPRESSION

Abstract
Two different chitosannanoparticals (CNP) samples were used as DNA carrier for transformation. Samples molecular weights weredetermined by viscometry method. Sample (A) was 501 kDa, which located in medium molecular weight chitosan range, while samples (B) was 827.9 kDa located in high molecular weight chitosan range. The degree of deacetylation (DDA) was determined by titration method.The sample(A) DDA was 66.26%, and sample (B) was 76.95%. CNP/DNA complex size were 109, 125 nm.,respectively.In case of E. coli DH5α normal cells (non-competent) the transformation efficiency of medium molecular weight CNP/DNA complex was3.50E+09, and2.14E+09for high molecular weight chitosanWhile no transformation in normal cells by using the same protocol dependent on heat shock.In case of competent cells, transformation efficiency was7.20E+11for CNPmedium molecular weight and 2.53E+10for CNPhigh molecular weight. While control transformation efficiencywas 4.48E+07.For Plant cells, the transformation percentage for medium molecular weight CNP was 32.24% and 14.57% forhigh molecular weight CNP.Plant single nodes were successfully transformedusing for both types of chitosan.
