The Role Of Cytokines In Experimentally Induced Rheumatoid Arthritis And Osteoarthritis In Temporomandibular Joint Of Guinea Pigs

Abstract


Much trials have been made to discover the underlying pathogenesis of osteoarthritis and rheumatoid arthritis. Recently many researches focused on the biochemical events associated with these diseases.
The objectives of this study were (a) TO investigate the role of cytokines in the pathogenesis of synovitis \Vhich occur during different stages of expenmentally induced RA and OA. (b) To correlate the clinical changes \\ h1ch occurred during different stages of experimentally induced RA and OA with the radiographic, macroscopic and microscopic changes.
Materials and Methods: This study was performed on one hundered adult guinea pigs. The animals were divided into five groups. In group I and II. RA was induced in the experimental animals by intraperitoneal injection (IP) of heat inactivated cells of group A streptococcus gyogcns. In group II the animals were injected intra-capsularly with recombinant Human IL-113 abut 1 ml immediately after induction of RA. In group III and group IV experimental osteoarthritis \vas induced in the experimental animals by extraction the posterior teeth unilateral!). In group IV the animals were injected intra-capsularly with recombinant Human IL-113. While group V was the control group . Five animals of each group were sacrificed by an over dose of ether at interval 48 hours, I ,6,12 weeks from the beginning of this study. Clinical examination, Measurement of the amount of IL-113. By using ELISA , Radiographic examination, Macroscopic examination and Micro- scopic examination were done.
Results: Progressive pathological changes were detected in RA with IL-113 and OA with IL-113 animal TMJ groups more than that in RA and OA animal TM.J groups. Clinical changes which occured during different stages of this study correlated with the radiographic, macroscopic and microscopic pathological changes.
Conclusion: IL-113 plays a role in the pathogenesis of OA and RA groups. IL-l [3 causes much more progressive pathologic changes in RA group than in OA group.
