UTILIZATION OF MODERN GENETIC TECHINQUES IN IMPROVEMENT OF EGYPTIAN RICE VARIETIES

Abstract
These studies were carried out in Rice Research and Training Center- Sakha - Kafr El-Shiekh, Genetics Dep. Labs- Faculty of Agric. Minufiya Univ., and The International Rice Research Institute (IRRI Philippines) during the years 1995-1998.
The studies included field and laboratory experiments and the goals ofthis studies were as follows:
1- To. determine the fingerprints for International and Egyptian rice varieties or lines by using PCR techniques.
2- To evaluate the parents for the restoration ability and high potassium affinity trait by using DNA markers.
3- To evaluate some of the Egyptian varieties and lines for the presence of
WCG.
4- To evaluate the parents for WCG by using the isozyme Amp/ which is tightly linked with the WCG.
I- Laboratory experiments:
Included the study of fingerprinting for 28 Egyptian rice varieties and lines plus 15 international lines by using 18 primers by PCR technique. Three types of prnners were found: primers which gave polymorphism before digestion, pruners showing polymorphism after digestion with restriction enzymes, and primers which did not show any polymorphism either before or after digestion. Data was analyzed by the computer program NTSYS to give the genetic distance between varieties and also to give the genetic tree.
The primer RG140 was used after digestion with EcoRl to study presence or absence of the Rf-3
