Taxonomic study in the Papilionoideae - Leguminosae

Abstract
Aims of the present study were improving the knowledge of taxonomic relationships of some closely related genera of the subfamily Papilionoideae; highlighting their similarities and close affinities; reassessment of their interrelationships based on previous and current systems of classification. This study comprised 36 taxa represented by 31 species; including five genotypes; belonging to 12 genera of the Papilionoideae that were either collected from the Egyptian floristic regions or provided by the Egyptian and German botanical gardens.
The study consulted five sources of taxonomic evidence in gaining the characters sets. Each of the following characters were recorded in the studied taxa: morphological features, the seed protein profiles produced by SDS-PAGE, molecular mass (Mr) in kilo-Daltons (kDa) of their polypeptide subunits, the Isozyme bands profile (Zymograms) of the enzymes -esterase (-Est), -esterase (-Est) and alcohol dehydrogenase (Adh); diploid and basic chromosome counts; ISSR-PCR fingerprints of DNA established by Five primers; HB09, HB10, HB13, HB14, HB15 and their molecular mass (MS) in base pairs (bp).
Taxonomic relationships were established by using UPGMA-clustering method based on each of 173, 97, 65, 20 & 178 characters of morphology, seed protein, isozymes, chromosome counts and ISSR, respectively; separately and in combination (533) and represented by phenograms. Each of the produced data sets was tested for consistency by measuring phenogram distortion from its relevant similarity matrix based on cophenetic (ultrametric) correlation coefficient (r). This coefficient was also used to compare the association of the data sets to each other. A principal components analysis (PCA) was also performed to discriminate the most important contributed characters in the clustering analysis. All analyses were carried out by using the program NTSYS-pc 2.2 (Rohlf, 2005).
The produced r values from comparison of the phenograms to each other indicated that, the combined data set exhibited the highest reliability and consistency among the used data sets. Thus, the produced classification based on the combined data been used to display and discuss the data of the present study.
The most important findings of the present study were:
1) Confirming the importance of integration of more than one source of taxonomic evidence in deepening the variation and increasing the differentiation between taxa, especially when includes the morphological characters.
2) Compatibly of both biochemical markers (SDS-PAGE & Isozymes) and molecular markers (ISSR) in revealing a high level of polymorphism among the studied taxa and thus, proved to be effective in differentiating taxa at the specific level and below.
3) To the best of our knowledge, the somatic chromosome count of 2n= 16 in Melilotus graecus (Boiss. & Spruner) Lassen has been recorded for the first time in the present study. Chromosome counts of the remaining taxa were compatible with their previous record and have been supported.
4) Confirming that x= 7 as the basic number in the genus Anthyllis in contrary to report of González (2004) and suggesting that, the other adverse counts could be just an aneuploid cytotypes of the genus.
