The Behavioural Response of Rhodobacter capsulatus B10 and 37b4 to Cadmium and Zinc toxicity

Abstract


This study is aiming to investigate the effect of Cd2+ (10 and 20 ppm) and Zn2+ (20 and 40 ppm) on growth , photopigments and morphology of R. capsulatus B10 and 37b4. The former strain has a capsule and the latter is capsuless and both strains belong to purple nonsulfur bacteria " Rhodospirillaceae". Particular attention was focused on bioaccumulation, cadmium- and zinc-binding proteins as well as the patterns of protein and lipopolysacchaddes by gel filtration and SDS-PAGE techniques.

1) Maximum growth of the two strains were reached after 4 days of incubation when they grown in RAH medium phototrophically under anaerobic conditions at 32 °C.
2) As the concentration of both cations increased in the growth medium, the maximum growth rate decreased, the mean generation time prolonged and tolerance index, protein content and bacteriochlorophyll-a content decreased. There were no qualitative changes in the position of absorption maxima of bacteriochlorophyll­ a and carotenoids, only a decrease in the absorption maxima.

R.capsulatus 37b4 grown at 20 ppm Cd2+.
