Differential Display Analysis of mRNA in Chronic Myelogenous Leukemia.

Abstract

Chronic myelogenous leukemia (CML) is characterized by a case of balanced reciprocal chromosomal translocation between chromosome number 9 and 22 which results in genesis of Philadelphia chromosome. About 95% of CML patients are characterized by Philadelphia chromosome, or at the molecular level by bcr-abl transcript (s). Hence the previous patients called Philadelphia positive (Ph+ve) and the rest called Philadelphia negative (Ph-ve) CML patients.

In this study, differential display analysis of Philadelphia positive and negative of CML showed that Ph+ve shared Ph-ve in some transcripts. Moreover, the Ph-ve patients expressed novel transcripts which were not observed in these of Ph+ve patients.

The chronic phase of CML represents the suitable phase of treatment and the changes from chronic to blast crisis phase which occur suddenly represent a dramatic problem because of the short survival of these patients. Differential display pattem or e()ch phase revealed that the pattern itself can be used as n new diagnostic marker for the blast crisis phase which is characterized by down regulation of most expressed genes in the chronic phase of CML.
