BIOLOGICAL ACTIVITY OF MONASCUS SPECIES ON SOME FOOD INDUSTRIAL WASTES

The biomass and pigment production by 3 strains of Monascus purpureus, and one isolate (which isolated from rice and identified as Monascus ruber strain based on 18s rRNA sequence) were studied under static and submerged conditions on Potato dextrose broth (PDB) and whey media. Whey medium was more effective on growth and pigment production than Potato dextrose broth (PDB) medium by all tested strains under liquid surface fermentation. Among Monascus species (3 strains and one isolate), M. purpureus DM1203 and M. ruber MI isolate were selected which gave the highest yield of pigment (72.6 & 67.5 U/L) and biomass (6.11 & 5.73 g/L). Optimized the pigment and monacolin productivity by selected Monascus strains on modified medium consisted of whey and okara using solid state fermentation at initial moisture content was 35% and 50% with pH 4.5 and incubated at 30°C for pigment production by M. purpureus DM1203 (385 U/L) and M. ruber MI (428U/L), respectively. Whereas, the maximum production of monacolin (11.82 and 14.16 µg ml-1) was found at 50% moisture level and pH 4.5 then incubated at 25°C and for M. purpureus DM1203 and M. ruber, respectively. Monacolin analyzed by HPLC was 14.92 and 24.31 µg ml-1 for M. purpureus DM1203 and M. ruber MI, respectively. Monascus fermented product was higher nutritive values of total protein, fats, ash and vitamins (B-complex), anti-oxidant activity. Moreover, it was found as anti-microbial effect against some pathogenic microorganisms and anti-cholesterol effect (which tested on rats in vivo).
