Abstract


In the present thesis a modem molecular biology study was carried out.


The objectives of this thesis were:


Isolation, characterization, cloning and determination of nucleotide sequence of the antibiotic resistance gene in Pseudomonas aeruginosa.


These objectives were achieved by the following:


• Isolation and identification of some pathogenic bacterial isolates of Pseudomonas aeruginosa. Identification was carried out using manual methods (including morphological, physiological and biochemical characters), molecular biology methods (including polymerase chain reaction (PCR) DNA fingerprint, electrophoretic separation of proteins (biochemical markers) and immunological methods using polyclonal antibodies.
• Screening the identified isolates for their resistance to different antibiotics and determination of MICs for the selected antibiotics against these organisms.
• Electrophoretic separation of general protein patterns of both Susceptible and resistant isolates which were put under antibiotic stress.
• Western blotting of protein patterns of both resistant and susceptible isolates.
• Development of anti OprM antiserum in white
Newzeland rabbits.
