Study of angiotensin converting enzyme gene polymorphism in preeclampsia
Essam Aly Galal;
Abstract
This work aimed at checking for the presence of Angiotensin Converting Enzyme gene polymorphism among some Egyptian women affected with PET, using accurate methods of molecular biology techniques as PCR for diagnosis and estimation of risk of the disease by comparing to normal population controls.
Studied women were represented by a case control study selected 72 Egyptian women at age ranging from 20 – 28years and mean was 24.1 years and SD + 2.03 years. 32 (44.4%) had a positive family history of PET, 41 (56.9%) were primigarvida, 31 (43.1%) were multigravida. For comparison, 94 healthy individual of nearly matched age and with no history of any cardiac diseases, DM, renal diseases, premature rupture of membranes or clinical chorioamnionitis and patients not in labour.
This women were taken from these presenting with PET to reception of Mansoura International Specialized Hospital, during the time from December 2008 to April 2009. These women were questioned about their history of hypertension, diabetes, and renal diseases and PET in first degree relative; smoking habits, and leisure time physical activity, Blood pressure, weight, height and waist and hip circumferencess were measured during a physical exam, and a blood sample was drawn to measure blood glucose, complete blood count, liver function, renal function, and ACE genotypes .
Three ml venous blood sample was collected from each subjects in a polyethylene tubes containing EDTA solution as an anticoagulant for DNA extraction. The Generation DNA Purification Capture Column Kit (Gentra systems USA) was used DNA extraction.
