GENETICS OF BREAST CANCER

HOSSAM HAMDY EL SAYED SHOAIB;

Abstract

reast cancer is a major cause of mortality in "11'-,lfemales all over the world. The genetic factor of the disease has been reported to have an
important role in the development of the disease. The principle genetic lesions observed include gene amplification, deletion, point mutation, loss of heterozygosity (LOH), and overall aneuploidy. Genes have been cloned (p53 and BRCAl) and another localized (BRCA2) whose function as tumour suppressor genes are compromised when inherited as mutant alleles. A progressive cascade of genetic changes occurs in the course of development of breast cancer. Successive generations of tumour cells overgrow and dominate the tissue, eventually giving rise to clonal or nearly clonal metastatic disease. Genetics of inherited breast cancer also is an important subject to study.
Finally, the most recent methods and strategies of biologic and gene therapy of breast cancer, antibodies and vaccines were also summarized in this essay.
