Molecular Approach for differentiation of some higher plants
Hala Mohamed Abdou El-Atroush;
Abstract
In the present work, the biochemical and molecular studies were used to document the existence of genetic markers that distinguish between two different plants namely, Medicago sativa and Medicago polymorpha. (1) Biochemical studies: Involved the spectrophotometery of the total protein concentrations. Simple protein, lipoprotein, glycoprotein and denaturated protein were studied by means of polyacrylamide gel electrophoresis (PAGE) technique for the different organs of the two plants (seeds, flowers, stems and leaves). (2) Molecular studies: a) The genomic DNA restriction
biochemical and molecular studies, it can be easier to
differentiate between the two plants under investigation (M.
sativa and M. polymorpha).
Key words: Medicago sativa - Medicago polymmpha Simple protein - Lipoprotein - glycoprotein - SDS protein - restriction endonuclease - DNA - RAPD - PCR.
enzyme pattern for M. sativa and M. Polymorpha was done
using ten restriction endonucleases to cut the genomic DNA of each plant and b) DNA fingerprinting by arbitrary primers/PCR amplification, twenty oligonucleotides of random sequences-10-mers-tested to amplified the DNA extracted from M. sativa and M polymorpha yang green leaves. From the twenty primers fifteen were selected.
On the basis of the results obtained from both the
biochemical and molecular studies, it can be easier to
differentiate between the two plants under investigation (M.
